I have a group registration set up for an event.  I have the main group member wanting to register the group but they will not be attending the conference.  Should I have register on behalf enabled on all paths?

Yes, enabling Register on Behalf on all paths would be the best approach. This allows the main group member to register others without being required to attend the conference themselves.
